2.3. Sampling ａnd chemical analyses 

The yields were five consecutive years (2014–2019), so we explained the mechanism of increasing yield of controlled-release potassium chloride through physiological ａnd biochemical indexes during growing seasons 2018? 2019. Samples from undisturbed surface soil (0–20 cm) were collected at seedling stage (31 days after fertilizing), re-greening stage (157 days after fertilizing), ａnd after dressing at jointing stage (188 days after fertilizing), grain-filling stage (210 days after fertilizing) ａnd at maturity (245 days after fertilizing). At jointing ａnd grain-filling stages, relative chlorophyll content (SPAD value) was determined by a chlorophyll meter (SPAD-502, Minolta, Japan) ａnd conducted by Li et al. (2019). The photosynthesis of the ear leaf was measured by a Li-6400XT portable photosynthesis system (Licor Inc., Lincoln, USA). The leaf area index (LAI) by a canopy analyzer (LAI-2200, Beijing, China) was evaluated at five different points. Leaf ａnd root samples were collected from the wheat during the jointing ａnd grain-filling growth stages. Auxin (IAA) ａnd abscisic acid (ABA) were estimated using the ELISA kit provided by Shanghai Hengyuan Biological 114 Technology Co. Ltd. (Shanghai, China). Soil samples of plowed horizons (0–20 cm) were collected from each plot, then air-dried in the laboratory, ａnd any visible roots, organic residues, ａnd stone fragments were removed manually, used by 2-mm, ａnd 0.25-mm sieves, ａnd further analyses. The available K of soil was extracted with 1.0 mol L? 1 of CH3COONH4 (7.0 pH) ａnd analyzed with a flame photometer (MODEL410, Sherwood, England).

Biochemical measurements

For Chlorophyll content measurement, six-month old leaves of wild type ａnd the mutant grown under identical conditions were collected ａnd Chl (a,b) were extracted in 90% acetone ａnd determined spectrophotometrically by the method of Lorenzen [26]. For the contents of chlorophyll intermediate products, leaves were homogenized in nine volume of 0.01 M Phosphate Buffered Saline (PBS), ａnd mixed on the ice ａnd centrifuged (30 min at 2,500g). The supernatants was assayed by a ELISA kit (HengYuan Biological Technology Co.,Ltd, Shanghai, China) separately. Data were analyzed by SPSS software.
